At puberty, male hormones stimulate growth of skin cells and increased production of sebum by sebaceous glands. The gland duct may become blocked by skin cells to form a whitehead, or by a plug of sebum that darkens on exposure to air, forming a blackhead, when sebum continues to be produced in a gland with a blocked duct, it may be forced out into surrounding tissue. Bacteria that are normally present in the skin (and harmless in other circumstances) cause the sebum to break down into chemicals that cause inflammation, resulting in the tender red lump we call a pimple. The pus formed by the inflammation usually pushes towards the surface from a 'head' that breaks through the blockage of the duct, and the pimple drains and disappears, leaving no scar.

Acne is also genetically influenced. If the parents have acne, then there kids are also more prone of developing acne. Use of generally cosmetics may also lead to occurrence of Acne.

Due to rubbing the skin, friction is caused which may cause acne. Various environmental factors such as pollution and high level of humidity also leads to formation of acne.

Acne is not caused by chocolate and greasy foods or stress.

If the duct is very tightly plugged, the inflammation may spread to the deeper layers of the skin, forming a 'blind' pimple. In some cases cysts form deep in the skin and raise purplish lumps that can take months or years to heal. Cystic acne is the most severe form, and almost always results in some pitting and scarring.

Why do some people get worse acne than others? Some people's sebaceous glands are extra sensitive to male hormones. This sensitivity seems to be inherited, explaining why acne tends to run in families. Acne in teenagers is rarely caused by an excessof male hormones, the skin is just over-reacting to the normal amount.

All forms of acne have their origin in wrong food habits, such as irregular hours of eating, improper food, excess of starch, sugar and fatty foods. Chronic constipation is another major cause of acne. If the bowels do not move properly, the waste matter is not eliminated as quickly as it should be and the blood stream becomes saturated with toxic matter. The extra efforts of the skin to eliminate this excess waste result in acne and other forms of skin disease. Yet another important cause of acne is a devitalised condition of the skin resulting from unhygenic living habits. Other contributing factors to the disorder are excessive use of tea, coffee, alcohol and tobacco, strenuous studies and sedentary habits which lead to indigestion and general debility.

Several factors are known to be linked to acne:

Accumulation of dead skin cells

Bacteria in the pores

Skin irritation or scratching of any sort

Hepatitis C virus (HCV) is the most common blood-borne infection in the United States, affecting about 1.3 percent of the population. Up to one-in-five sufferers develop liver cirrhosis, and up to one-in-20 develop liver cancer. HCV is the top reason for liver transplantation, and the 16th leading cause of premature death in the country. Recent evidence has suggested that disease burden and mortality from chronic HCV infection may increase in the coming years, as the number of persons with longstanding infections continues to rise.

To update estimates of trends and demographics of hepatitis C-related mortality in the U.S., a team of researchers led by Matthew Wise of UCLA and including researchers from the CDC and the Los Angeles County Department of Public Health analyzed mortality rates derived from U.S. Census and multiple-cause-of-death data from 1995-2004. They included 56,409 HCV related deaths, including those for which the disease was the underlying cause; those for which chronic liver disease was the underlying cause and hepatitis C was a contributing cause; and those for which HIV was the underlying cause and chronic liver disease and hepatitis C were contributing causes.

During the study period, HCV-related mortality rates increased from 1.09 deaths per 100,000 persons in 1995 to 2.57 per 100,000 in 2002, before declining slightly to 2.44 per 100,000 in 2004. Average annual increases were smaller during 2000-2004 than 1995-1999. The most dramatic age-specific increases were observed among 45 to 54 year olds who had an increase of 376 percent, and 55 to 64 year olds who had an increase of 188 percent. For the latter group, rates rose for the entire duration of the study.

“The highest mortality rates were observed among males, persons aged 45-54 and 55-64 years, Hispanics, non-Hispanic blacks and non-Hispanic Native American/Alaska Natives,” the authors report. They suggest that demographic differences are related to prevalence among the various populations.

The observed increases likely reflect both true increases in mortality and the growing use of serologic tests for HCV, the authors say. “As such, true increases in hepatitis C-related mortality during 1995-1999 were likely more gradual than the observed trends, and differences in mortality patterns between the time periods are difficult to interpret.” While the study was limited by possible inaccuracies in death certificate data, the authors believe that this more likely lead to an underestimate of the true number of hepatitis C-related deaths.

“In summary, substantial increases in overall hepatitis-C-related mortality rates have occurred since 1995,” the authors conclude. “The relatively young age of persons dying from hepatitis C-related liver disease has made hepatitis C-related disease a leading infectious cause of years of potential life lost as well as an important cause of premature mortality overall.” They point out the ongoing need for measures to prevent progression of liver disease among those infected with HCV, and the need for ongoing analysis of mortality trends.

Preventing Spread Of Ovarian Cancer

A drug that blocks production of an enzyme that enables ovarian cancer to gain a foothold in a new site can slow the spread of the disease and prolong survival in mice, according to a study by researchers from the University of Chicago Medical Center, but only if the drug is given early in the disease process.

In the April issue of the Journal of Clinical Investigation, the researchers show that an enzyme known as MMP-2 is necessary for ovarian cancer to attach itself to the sites where it tends to spread. Several drugs known as MMP inhibitors (for example, marimastat or prinomastat) inhibit the enzyme, dramatically reducing the tumor's ability to establish itself at sites beyond the ovary. But such MMP inhibitors, which were abandoned after they failed to extend survival in earlier clinical trials, have to be given before the cancer has spread.

"Our study suggests that MMP-2 inhibitors could have a significant impact on ovarian cancer but only if administered quite early, before the cancer has advanced beyond the ovary," said Ernst Lengyel, assistant professor of obstetrics and gynecology at the University of Chicago.

This approach could help women who receive surgical treatment while the disease is still limited to the ovary as well as those who have successful surgery to remove all evidence of local spread of the disease. In the earlier trial, marimastat was given to women with late-stage disease that had already spread.

The fifth leading cause of cancer death in women, ovarian cancer -- unlike breast, colon or lung cancer -- tends to spread within the abdominal cavity and not to distant organs. Carried by fluid, it most often spreads throughout the peritoneal cavity and to the omentum, a large fat pad draped over the small bowel.

Lengyel and colleagues wanted to understand the many steps required for ovarian cancer to dislodge from its original site and establish itself elsewhere in the peritoneal cavity. They found that one of the key steps was production of MMP-2 by cancer cells that came in contact with the cells that line the peritoneal cavity.

When ovarian cancer cells make contact with the cells that line this internal cavity, they produce MMP-2 (an acronym for matrix metalloproteinase-2). MMP-2 alters two proteins--vitronectin and fibronectin--found on the surface of the cells that line the cavity. These alterations change those proteins in a way that enables the cancer cells to latch on to them better. Once attached, the cancer cells can multiply rapidly and invade.

By inhibiting MMP-2 activity early in the disease course, Lengyel and colleagues were able to prevent injected ovarian cancer cells from attaching to their target tissues in the peritoneum and omentum. This reduced the growth of new tumors by 68 percent, when measured four weeks after treatment.

The inhibitor nearly doubled survival time in mice that were injected with ovarian cancer cells. Those who received it survived an average of 63 days, compared to untreated mice, who survived only 36 days.

Brief and early intraperitoneal treatment with an MMP inhibitor, the authors conclude, may reduce peritoneal attachment, reduce metastases and significantly prolong survival.

The treatment has much less impact, however, once cancerous cells have attached and formed colonies. In several earlier trials, marimastat, an oral MMP inhibitor, was given for a prolonged period of time to women with late-stage disease that had already spread.

"MMP-inhibitors were given at the wrong time for too long, causing side effects," Lengyel said. Attachment is the first step for metastatic spread. MMP-2, the target of MMP inhibitors, plays a role in early cancer spread.

"Our study examines the initial step of ovarian cancer metastasis," the authors note, when cancer cells meet unprepared target cells. Other steps in this process, they suggest, may also provide additional treatment targets.
